The normal metabolism of amino acids is an important basis for life activities. All tissue cells can metabolize amino acids, including deamination, decarboxylation, ammonia metabolism and oxidative decomposition capacity.
Elabscience® offers a variety of methods to detect total protein content, such as BCA (E-BC-K318), Biuret (E-BC-K165), and Coomassie brilliant Blue (E-BC-K168). The biuret method was preferred to detect the total protein content in serum. If the sample contained chelating agent such as EGTA, high concentration EDTA or reducing agent such as DTT, mercaptoethanol, only Coomassie brilliant blue method was suggested for the detection. The BCA method was preferred to detect the total protein content in plant samples.

Appendix: Summary of total protein content determination methods
1． Easy operation, Microplate Reader is used.
2． High sensitivity, various sample types and less sample consumption.
3． It is not affected by the chemicals such as decontamination agents in most samples, and is compatible with the 5% TritonX-100, 5% SDS, 5% Tween 20,60,80 in the sample.
4． High accuracy. The coefficient of variation of different protein molecules was much less than that of Coomassie brilliant method.
1． Reaction time is a litter longer.
2． The result can be affected by the chelating agent and slightly high concentration reducing agent. It is necessary to ensure that EDTA concentration is less than 10 mM, no EGTA, DTT concentration is less than 1 mM, mercaptoethanol concentration is less than 1 mM.
Coomassie bright blue Method
1． High sensitivity.
2． The detection is rapid and simple. Short reaction time and good color stability, which can keep stable in 1 hour.
3． Less interfering substances, not affected by reducing agents such as mercaptoethanol, EDTA complexing agents, sugars, etc.
1. Because of the different contents of arginine and aromatic amino acids in various proteins, there will be a large deviation in the determination of different proteins for Coomassie brilliant blue method.
2. Decontamination agents such as Triton X-100, sodium dodecyl sulfate (SDS) and NaOH of 0.1m may interfere the determination of Coomassie brilliant blue.
1. The color of different protein produced by biuret method is similar in depth and light, and is not affected by temperature.
2. The detection is rapid and simple for detection of protein content and simple with single reagent.
There are many interfering substances, chelating agent and reducing agent will interfere with the reaction. Biuret reaction not only take places in protein, but also in molecules with two or more molecular structural formula.
