J Tian et al investigate the cadmium accumulation regulator and the harmful to host plant and leaf bacteria.. Peroxidase (POD) avtivity in crop leaf was determined using peroxidase activity assay kit (E-BC-K227-S).
It is recommended to take 2~3 samples with expected large difference to do pre-experiment before formal experiment and dilute the sample according to the result of the pre-experiment and the detection range (0.5-40 U/mL).
The recommended dilution factor for different samples is as follows (for reference only):

Asked: I have a problem with using this kit. I want to determine POD activity in tomato leaves. Tissue extract is 50 mg in 0,45 ml PBS with protein content about 1.5-2 mg/ml. I dilute the extract 1, 5, 10 and 20-fold (100 mcl extract, 20 extract + 80 PBS, etc.) and take into account dilution factor (f in the formula) - 1, 5, 10 and 20, respectively. The problem is that OD420 values does not change properly - it declines from 0,31 to 0,25 in controls and from 0,7 to 0,57 in samples. Using the formula from manual I get increasing results of POD activity with declining extract concentration - 24,9 U/mgprot in sample without dilution and 430 U/mgprot in sample with dilution factor 20. Could you please give me some hint? Maybe I do something wrong in my calculations?

Asked: Would this kit work for rose petals? It may produce some reddish purple extract, I found the detection wavelehgth is 420 nm, will it interfered?
