This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse IgG. Samples (or Standards) are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse IgG and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse IgG, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 ± 2 nm. The OD value is proportional to the concentration of Mouse IgG. You can calculate the concentration of Mouse IgG in the samples by comparing the OD of the samples to the standard curve.
Human immunoglobulin G (IgG) is the highest content of immunoglobulin in serum, accounting for approximately 75% of the total immunoglobulin content in plasma. There are four subtypes of IgG, namely IgG1, IgG2, IgG3, and IgG4, which each have different biological functions and characteristics. IgG is mainly used as an antibody for antiviral, neutralizing, antibacterial, and immune regulation in the body, and is also the only antibody that can be transmitted to newborns through the placenta. In addition, IgG is also involved in the development of autoimmune diseases, such as macrophages and neutrophils with IgGFc receptors on their surfaces, which can enhance their ability to phagocytose substances such as bacteria. IgG originates from human serum and has different molecular structures and biological characteristics. Its main biological functions include binding complement and Fc receptors, thereby activating a series of biological effects. These closely related glycoproteins are composed of 82% to 96% protein and 4% to 18% carbohydrates. In summary, human immunoglobulin G (IgG) is an important immunoglobulin that exists on the surface of blood and lymphocytes, has multiple biological functions, serves as the material basis for antibodies, and plays a crucial role in the human immune response.
