This ELISA kit uses the Competitive-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with Rat DHEA-S. During the reaction, Rat DHEA-S in the sample or standard competes with a fixed amount of Rat DHEA-S on the solid phase supporter for sites on the Biotinylated Detection Ab specific to Rat DHEA-S. Excess conjugate and unbound sample or standard are washed away, and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns from blue to yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of Rat DHEA-S in tested samples can be calculated by comparing the OD of the samples to the standard curve.
Dehydroisoandrosterone sulfate (DHEAS) is a chemical substance with the formula C19H28O5S. This compound mainly originates from the adrenal gland, especially through 17 β- The action of hydroxyl dehydrogenase and 5,4-disaminase produces testosterone. In the human body, almost all DHEAS comes from the adrenal gland, so it can be used as an indicator to evaluate adrenal function. In addition, the quantitative detection of DHEAS is of great significance for the auxiliary diagnosis of precocious puberty, female hirsutism, and polycystic ovary syndrome.
