The total price will increase by $120 when the quantity of marked antibody
increase by 1mg.
In this case, customer provides purified antigen, we prepare antibody and identified it by western blot, then determine titer by ELISA and test concentration by BCA.
Result
1. Result of Western blot
As the expression information of target protein was not told, we identified the antibody with immunogen. The sample amount is 1.5 μg.
The main band was at the position of 55~72 kDa and a few unpurposed bands showed up. That may due to some unpurposed protein exist among the antigenic protein.
2. Result of ELISA
The coated antigen was 0.2 μg. We detected antisera and purified antibody respectively with dilution factor 8×104, 16×104, 32×104, 64×104, 128×104, 256×104, 512×104, 512×104.The 3 times of the blank OD value was regarded as the titer of the antibody.

The titer of rabbit A is 128×104. The titer of rabbit B is 256×104.
The calculated concentration of rabbit A is 24 mg/mL.
The calculated concentration of rabbit B is 29.9 mg/mL.
3. Result of BCA
Dilute BSA with TE to constitute concentration gradient 0 mg/mL, 0.2 mg/mL, 0.4 mg/mL, 0.8 mg/mL, 1 mg/mL. Do duplicate and make standard curve. Dilute purified IgG 10 times, 20 times, 40 times with TE. Do duplicate (The volume of BCA is 200 μL, the volume of protein is 20 μL), keep at 37℃ for 30 min, read at 568 nm.
