Detection principle

Bromocresol green (BCG) is widely used as protein staining agent. BCG can combine the albumin in pH 4.0~4.2 to form an albumin-BCG complex. And the color changed from yellow to green. The depth of color is proportional to the concentration of albumin. The content of albumin in serum can be calculated indirectly by measuring the OD value at 630 nm.

ALB level was significantly induced in STZ treated group.

A A-R Mohamed et al found that selenium nanoparticle can alleviate cardio-hepatic damage in type II diabetes. Albumin (ALB) level in rat serum was determined using ALB colorimetric assay kit (E-BC-K057-M).

Dilution of sample

It is recommended to take 2~3 samples with expected large difference to do pre-experiment before formal experiment and dilute the sample according to the result of the pre-experiment and the detection range (0.08-15 g/L).

The recommended dilution factor for different samples is as follows (for reference only):

Sample type

Dilution factor

Human serum

Human plasma

HepG2 supernatant

Mouse plasma

Rat serum

Note: The diluent is normal saline (0.9% NaCl) or PBS (0.01 M, pH 7.4).

  • Application:Biochemical
  • Description:When I first saw this kit, I was attracted by the time required by the whole experiment process, and bought it with the mentality of trying it out. The result was in line with my expectation,but,It\'s a little tricky to do the standard curve,i still want to have a single standard to do the calculation.

Asked: Hello, I am looking into developing a simple positive identification in my sample. I basically need an assay that will show if albumin is present or absent, I don\'t need to find the concentration as long as their a big difference in the OD values between the (background) and the reference standard. Do you think that the Albumin (ALB) Colorimetric Assay Kit (Bromocresol Green Method) fits the job? if not, what would you reccommend
