Answered: The following are the detailed steps of how the cell samples were processed and how the protein concentration was determined. ① Harvest the number of cells needed for each assay (initial recommendation =3 × 10^6 cells). ② Wash cells with 0.9% NaCl . ③ Homogenize 3 × 10^6 cells in 200-400 µL 0.9% NaCl (μL) with a ultrasonic cell disruptor at 4℃. ④ Centrifuge at 10000 × g for 10 minutes to remove insoluble material. Collect supernatant and keep it on ice for detection. ⑤ Meanwhile, determine the protein concentration of supernatant (E-BC-K318-M).

ALP activity in Serum (plasma) sample Definition: The amount of 1 μmol p-nitrophenol produced by 1 L serum (plasma) per minute catalyzing the substrate at 37℃ is defined as 1 activity unit. Calculation formula: ALP activity (U/L) = (ΔA-b) ÷ a ÷ T × f 2. ALP activity in the tissue sample Definition: The amount of 1 μmol p-nitrophenol produced by 1 g tissue protein per minute atalyzing the substrate at 37℃ is defined as 1 activity unit.
