Application This ELISA kit applies to the in vitro quantitative determination of Human F9 concentrations in serum, plasma and other biological fluids.

Test Principle

This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human F9. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human F9 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human F9, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human F9. You can calculate the concentration of Human F9 in the samples by comparing the OD of the samples to the standard curve.

The recovery of Human F9 spiked at three different levels in samples throughout the range of the assay was evaluated in various matrices.

Assay Procedures

1. Add 100μL standard or sample to the wells. Incubate for 90 min at 37°C

2. Discard the liquid, immediately add 100μL Biotinylated Detection Ab working solution to each well. Incubate for 60 min at 37°C

3. Aspirate and wash the plate for 3 times

4. Add 100μL HRP conjugate working solution. Incubate for 30 min at 37°C. Aspirate and wash the plate for 5 times

Asked: I have some questions related to storage of human F9 ELISA Kit (E-EL-H0760). In your instruction, Kit can be stored in 4 C when kit is used within 1 month. Otherwise, components inside kit should be stored separately 4 C and -20 C. But one of my colleagues, had some mistakes. The kit have been remained unopened and stored in 4 C for 28 days (received at Jan 24, 2024; today is Feb 22, 2024). In this case, we wonder whether the kit can be used after transfer of some components to -20 C even after 1 month. Please reply to me asap after checking this Q&A. Many thanks for your answer in advance.

Answered: Please confirm whether the kit can be used for more than a month even if it is stored at 4℃. ---The kit can be stored at 2-8℃ for 3 months in the case of unopened. If the kit is used after 3 months, store the items separately according to the manual once the kit is received. If I store a component at 4℃ for 4 weeks and then transfer it to -20℃, how long can I still use the kit? ---5 months.
