The level of MDA was induced significantly in treated cells. (***P<0.001)
H Huang et al developed a nanoplatform that can induce ferroptosis for cancer therapy. Malondialdehyde (MDA) CAL-27 cell was determined using MDA colorimetric assay kit (E-BC-K028-M).
It is recommended to take 2~3 samples with expected large difference to do pre-experiment before formal experiment and dilute the sample according to the result of the pre-experiment and the detection range (0.29-100 nmol/mL).

Answered: Yeah, for E-BC-K028-M kit, the Bradfrod method can also be used to detect protein concentration.

Asked: Hi, I was wondering if this kit can be used for supernatant from mouse skin? My samples are mouse skin, which I homogenise in RIPA and centrifuge. The resulting supernatant is what I will use for the assay.

Answered: 1. E-BC-K028-M is suitable for the detection of cell samples. If the MDA content of tissue samples is detected, it is recommended that you take a look at the E-BC-K025-M kit. 2. Since the detection principle of E-BC-K025-M kit is chemical reaction, the composition of RIPA cracking solution is relatively complex, which may have an impact on the reaction system. It is not recommended to treat samples with RIPA lysate. 3. If testing tissue samples, sample preparation is recommended in accordance with the manual. For skin tissue, it is recommended to extend the homogenate time until the tissue is fully broken.

Asked: Hi, I\'m using HaCaT cells. Firstly, may I know if I can use RIPA buffer instead of the reagent 5 (extraction solution) in the E-BC-K028-M kit? Secondly, does using Trypsin for cell detachment is contraindicated even if I rinse twice with PBS?

Answered: Dear Elly, Regards to your questions, please refer to the feedbacks below: 1. Since E-BC-K028-M kit is developed on the principle of chemical reaction, the complex composition of RIPA buffer may have an impact on the reaction system. It is not recommended to replace reagent 5 with RIPA buffer to treat cells. 2. If it's to detect the adherent cells, the cells can be digested with trypsin or scraped off with cell scraper. If it's digested with trypsin, it needs to wash with PBS for 2~3 times. After that it would not affect the result. Hope it helps Best Regards Lizzy

Asked: Hi, can I know if reagent 5 is the extraction solution? Meaning that we don’t have to use other homogenization medium such as Triton-X? I tried few times and the standard is the only group that gives me colour.

Answered: Hi Nabila. Yes. We suggest to increase the concentration of cell homogenization. And try to read the OD value, the color might be quite light.
