Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to MIP-3α . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for MIP-3α and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain MIP-3α , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 44 nm. The OD value is proportional to the concentration of MIP-3α . You can calculate the concentration of MIP-3α in the samples by comparing the OD of the samples to the standard curve.
Background
Chemokine ligand 20 (CCL20), also known as macrophage inflammatory protein 3α (MIP-3α), is a small molecular weight cytokine in the CC chemokine family. CCL20 strongly chemotaxis lymphocytes and weakly neutrophils. CCL20 can be expressed in monocytes, macrophages, microglia, astrocytes, mast cells, keratinocytes, intestinal epithelial cells, osteoblasts, fibroblasts, endothelial cells and bronchial epithelial cells. CCL20 chemotaxis lymphocytes and dendritic cells to epithelial cells surrounding mucosal lymphoid tissue, thus participating in the formation and function of mucosal lymphoid tissue. CCL20 binds and activates the chemokine receptor CCR6 to target cells. CCL20 may play an important role in the pathogenesis of autoimmune diseases in the central nervous system.
