Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to CX3CL1 . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for CX3CL1 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain CX3CL1 , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 51 nm. The OD value is proportional to the concentration of CX3CL1 . You can calculate the concentration of CX3CL1 in the samples by comparing the OD of the samples to the standard curve.
Background
CX3CL1 is a 373-amino acid macrocytokine protein that contains multiple domains and is the only known member of the CX3C chemokine family.It is also commonly referred to as fractalkine (in humans) and neurotactin (in mice).The polypeptide structure of CX3CL1 is different from the typical structure of other chemokines.For example, the spacing of characteristic n-terminal cysteine is different;There are three amino acids in CX3CL1 that distinguish the initial cysteine, none in the CC chemokines, and only one intermediate amino acid in the CXC chemokines.CX3CL1 is a long protein (373 amino acids in the human body) with an extended mucin-like stem and chemokine domain.Soluble CX3CL1 efficiently chemically adsorb T cells and monocytes, while the cells bind chemokines to promote the strong adhesion of leukocytes to activated endothelial cells, in which it is mainly expressed.
