X Zhang et al investigate the usage of H2O2 and O2 for cooperative chemodynamic/starvation cancer therapy. Alanine aminotransferase (ALT) avtivity in mouse blood was determined using ALT activity assay kit (E-BC-K235-M).
It is recommended to take 2~3 samples with expected large difference to do pre-experiment before formal experiment and dilute the sample according to the result of the pre-experiment and the detection range (0.75-72.3 IU/L).
The recommended dilution factor for different samples is as follows (for reference only):
Sample type |
Dilution factor |
Human serum |
1 |
Human plasma |
1 |
Porcine serum |

Rat serum |

10% Rat liver tissue homogenization |

10% Rat kidney tissue homogenization |

Note: The diluent is normal saline (0.9% NaCl) or PBS (0.01 M, pH 7.4).

Asked: Can you please help me understand exactly what a carmen unit is and why we are using it instead of the conventional method of plotting a standard curve of concentration of standard vs the absorbance. How is the carmen unit relevant to this exact calculation I would also like to know how you arrived at the numbers 0, 28, 57, 97, 150, 200 for the standard Carmen values.

Answered: 1. This indicator is special, and the method used is the Reitman-Frankel method. The standard curve of the Reitman-Frankel method is determined by comparing the colorimetric method experimental result with the result of karmen spectrophotometry (rate method), and the report result of Karmen units will be more accurate. 2. The Karmen's units corresponding to the standard are fixed and converted according to the concentration of pyruvate.
