Plasma D-LA level was significantly increased in VDN group, and reversed in SP treated group. (****P<0.0001)
Jianlong Yan et al investigates the beneficial effect of the short-chain fatty acid in intestinal microbiota remodeling. Plasma D-Lactate of rat was determined using D-lactate colorimetric assay kit (E-BC-K002-M).
It is recommended to take 2~3 samples with expected large difference to do pre-experiment before formal experiment and dilute the sample according to the result of the pre-experiment and the detection range (0.06-8.0 mmol/L).

Asked: Must I use 530 nm for the detection? I only have a microplate reader with 540 nm.

Answered: 540 nm also works for this assay. Actually, the range between 520-550 nm is working, while 530 is the best choice with highest sensitivity.

Answered: It is not recommended to use lysate. Some components of lysate may interfere with the detection. Biochemical kits generally use the recommended homogenization medium for ultrasonication or mechanical homogenization.
