Answered: EL-R0015 kit is suitable for the detection of rat brain tissue samples. We have verified normal rat whole brain tissue undilution and the sample measured value was 20-100pg/mL. However, there are individual differences between samples, so it is suggested that you can confirm it through pre-experiment. In addition, it is not recommended to use special lysates when collecting tissue samples. In generall, the tissue samples can be collected as follows for ELISA assays. 1) Mince the tissues to small pieces as much as you can and rinse them in ice-cold PBS (0.01M, pH=7.4) to remove excess blood thoroughly. 2) Tissue pieces should be weighed and then homogenized in PBS (tissue weight (g): PBS volume (mL) =1:9) with a glass homogenizer on ice(It's suggested to add an appropriate amount of protease inhibitor to PBS before the homogenization). 3) To further break the cells, you can sonicate the suspension with an ultrasonic cell disrupter or subject it to freeze-thaw cycles. 4) The homogenates are then centrifuged for 5 minutes at 5000×g at 2 - 8℃, collect the supernatant to carry out the assay. To get more accurate result, it's suggested to run the assay with freshly collected samples.
