Detection Principle Acetate kinase (AcK) is a member of the ASKHA (acetate and sugar kinases, Hsp70, actin) phosphotransferase superfamily and serves as a central enzyme in carbon and energy metabolism in prokaryotes.
AcK catalyzes the acetate reaction with concomitant consumption of NADH. NADH exhibits a maximum absorption peak at 340 nm, and AcK activity is calculated by measuring the rate of decrease in absorbance at 340 nm.
