Precision

Intra-assay Precision (Within-run Precision): Three samples representing low, mid, and high concentrations of Human IL-6 were tested 20 times on a single plate.

Recovery

Linearity

Linearity of the assay was evaluated by spiking samples with high concentrations of Human TAT and performing serial dilutions using Standard & Sample Diluent to produce concentrations spanning the assay's dynamic range. Test Principle

This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human TAT. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human TAT and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human TAT, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human TAT. You can calculate the concentration of Human TAT in the samples by comparing the OD of the samples to the standard curve.

Background

Thrombin, which cleaves bonds after Arg and Lys, converts fibrinogen to fibrin and activates factors V, VII, VIII, XIII, and, in complex with thrombomodulin, protein C. Functions in blood homeostasis, inflammation and wound healing. Antithrombin: Most important serine protease inhibitor in plasma that regulates the blood coagulation cascade. Antithrombin inhibits thrombin, matriptase-3无TMPRSS7, as well as factors IXa, Xa and XIa. Its inhibitory activity is greatly enhanced in the presence of heparin.
