01 Master the Core Four Steps to Nail Most ELISAs
No matter which type of ELISA kit you use, the core principles and workflow are similar. Mastering the following key points is essential for obtaining reliable data.
1. Preparation and Sample Addition
Reagents to Room Temperature: Always bring all reagents to room temperature before use.
Standards are Key: Reconstitute and dilute standards exactly as instructed. Always change pipette tips between dilutions.
Slow Addition Along the Well Wall: Add samples slowly along the bottom of the well wall to avoid bubbles and splashing.
Cover and Incubate: Seal the plate with sealing film and incubate for the specified time at the required temperature.
Wash Thoroughly: For manual washing, we recommend: fill wells completely with wash buffer, let stand for 60 seconds, aspirate and flick, and repeat 3 times. After the final wash, blot the plate firmly onto absorbent paper to remove residual liquid.
3. Color Development and Stop
Develop in the Dark: Add TMB substrate and develop protected from light. Stop the reaction once a clear blue gradient is visible.
