01 Detection Principle of the Cell Viability Detection Product

This product enables cell viability detection based on the ATP luminescence method. The core mechanism is as follows: ATP is a key energy metabolite in oxidative phosphorylation, and changes in its concentration reflect changes in cell viability.

Released ATP acts as a specific substrate for luciferase, triggering a cascade of enzymatic reactions with luciferase and luciferin. This reaction produces a chemiluminescent signal that can be captured by a reader. The luminescent signal shows an excellent linear relationship with ATP concentration in the sample, which directly corresponds to the number of viable cells before lysis. Thus, by detecting the luminescence intensity, cell viability can be accurately quantified.

Hela cells treated with 0.2 μM and 2 μM antimycin A for 1 hour showed expected trends in cell viability compared to untreated cells when measured with Cell Viability Chemiluminescence Assay Kit (E-BC-F200).
