Detection principle

AST/GOT enables alpha-ketoglutaric acid and aspartic acid to displace amino and keto groups to form glutamic acid and oxaloacetic acid. Oxaloacetic acid can decarboxylate itself to form Pyroracemic acid during the reaction. Pyroracemic acid reacted with 2,4-dinitrophenylhydrazine(DNPH) to form 2,4, dinitrophenylhydrazone showing reddish brown in alkaline solution. Measure the OD values and calculate the enzyme activity.

No significant difference of AST activity was observed between WT and 3SA mice after HFD for 17 weeks.

H Xia et al investigate the relationship between insulin resistance and GSK3β-FBXW7-ERRα transcriptional axis. Aspartate aminotransferase (AST) activity in mouse serum was determined using AST activity assay kit (E-BC-K236-M).

Dilution of sample

It is recommended to take 2~3 samples with expected large difference to do pre-experiment before formal experiment and dilute the sample according to the result of the formal experiment and the detection range (1.1-72.3 IU/L).

Asked: Is it possible to perform this AST activity assay on frozen samples. For example if I have done the sample preparation on mouse livers, if I store the extracted samples at -80°C for a few nights can I reuse those same samples for the enzyme activity test?

Answered: If you has no time to test on the same day, the tissue can be aliquoted and frozen at -80 degrees. If the supernatant after homogenization is processed, it is not recommended to test overnight, which will affect the enzyme activity. The tissue homogenization can be performed when you needs to test.

Asked: I want to know what is the exact difference between the sample and the control reactions. Both wells have all the same reagents added to them and also both have the sample. The sample is just added at different times, does this cause a change in the reaction?

Answered: Set the control is to exclude the interference of pyruvate and non-enzymatic reaction of the sample. Reagent 4 has an inhibitory effect on the enzyme and can terminate the reaction between substrate and enzyme.
