Linearity
Linearity of the assay was evaluated by spiking samples with high concentrations of Mouse TSH and performing serial dilutions using Standard & Sample Diluent to produce concentrations spanning the assay's dynamic range. This CLIA kit uses the Sandwich-CLIA principle. The micro CLIA plate provided in this kit has been pre-coated with an antibody specific to Mouse TSH. Samples or Standards are added to the micro CLIA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse TSH and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse TSH, biotinylated detection antibody and Avidin-HRP conjugate will appear luminescence. The Relative light unit (RLU) value is measured by the Chemiluminescence immunoassay analyzer at 425nm. The RLU value is positively associated with the concentration of Mouse TSH. You can calculate the concentration of Mouse TSH in the samples by comparing the RLU value of the samples to the standard curve.
Thyrotropin releasing hormone (TRH) is a tripeptide hormone that stimulates the release of thyroid stimulating hormone and prolactin by the anterior pituitary. TRH is produced by the hypothalamus and travels across the median eminence to the pituitary via the hypophyseal portal system. In addition to the brain, TRH can also be detected in other areas of the body including the gastrointestinal system and pancreatic islets.
