Background

Tetracyclines are broad-spectrum antibiotics, among which oxytetracycline and doxycycline are the most common veterinary drugs. In addition to therapeutic use, in many other countries, tetracycline is usually added to livestock feed in sub-therapeutic doses as a growth promoter for pigs, poultry, and aquaculture. Excessive use leads to the emergence of antibiotic resistance and allergic reactions.

Asked: Dear Sir or Madam, We aim to use the TCs (Tetracyclines) ELISA Kit to detect the tetracycline in Drosophila transgenic flies (females) and their eggs. For this purpose, the wild type female flies that fed with or without tetracycline can be used as positive and negative controls, respectively. The user manual shows that this kit can be used for tissue and eggs. We are wondering if additional controls are also needed for eggs. For example, would egg yolk can interfere with ELISA assays, or both tissue and eggs respond to this assay in a same way? For either case, could you please provide some supporting information such as technical data or relevant publications. Thanks you very much!

Answered: Dear Ying Thanks for your mail. For the requested item E-FS-E064 TCs (Tetracyclines)ELISA Kit 1. It can be detected, but pre-experiments are required to verify whether the kit is suitable for Drosophila muscle samples. Usually the test sample is the muscle tissue of livestock (chicken, duck, pig and other edible animals) or eggs (Edible eggs are not egg cells), and There is no related experiments done with Drosophila. It is recommended to use "3.1 Pretreatment of muscle (method 1) (livestock) sample" in E-FS-E064 to test the content of tetracycline in Drosophila muscle. Please note that the sampling amount of the same sample has to be exact within 1±0.01g. Note: Drosophila has a certain ability to absorb and metabolize drugs. A small amount of tetracycline drugs are metabolized and will not remain in the muscle tissue of Drosophila. The final test results are the same as those of Drosophila without drugs. 2. Usually tetracycline drugs that are not fully metabolized in Drosophila remain in muscle tissue and possibly in Drosophila eggs. But we can not guarantee that it will be present in the egg cell, and testing Drosophila eggs is not recommended because the positive sample is not available 3. Relevant literature research cannot be provided, but if you are interested, we can provide a test report on muscle samples for E-FS-E064 kit validation is provided. Best regards
