Precision

Intra-assay Precision (Within-run Precision): Three samples representing low, mid, and high concentrations of Human IL-6 were tested 20 times on a single plate.

Recovery

The recovery of Mouse CCL1 was evaluated by spiking samples at low, mid, and high concentrations across the assay range in various sample matrices.
The assay performance was assessed by comparing the measured concentrations to the expected spiked amounts to determine the percent recovery.

Linearity

Linearity of the assay was evaluated by spiking samples with high concentrations of Mouse CCL1 and performing serial dilutions using Standard & Sample Diluent to produce concentrations spanning the assay's dynamic range. The measured values were then compared to the expected concentrations to assess the linearity of response.

For the recovery experiment, the samples were spiked with standard solutions of a specific concentration and then serially diluted to 2-fold, 4-fold, 8-fold, and 16-fold dilutions, respectively. The average recovery rate was subsequently calculated.

Test Principle

This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse CCL1. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse CCL1 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse CCL1, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 5 nm. The OD value is proportional to the concentration of Mouse CCL1. You can calculate the concentration of Mouse CCL1 in the samples by comparing the OD of the samples to the standard curve.

Background

Mouse CCL1 also known as T-cell activation protein 3 or TCA-3 is a member of the CC chemokine family. This cytokine is chemotactic for monocytes but not for neutrophils. It acts by binding to the seven transmembrane spanning G-protein-coupled receptor CCR8. CCL1 mediates inflammatory immune responses, viral infections, and tumorigenesis by attracting monocytes, natural killer cells, and immature B-cell nuclear dendritic cells. It has also been shown to chemoattract T-cells, contributing to the inflammatory response through leukocyte recruitment.
