Recovery
The recovery of Mouse NOV was evaluated by spiking samples at low, mid, and high concentrations across the assay range in various sample matrices.

Linearity
Linearity of the assay was evaluated by spiking samples with high concentrations of Mouse NOV and performing serial dilutions using Standard & Sample Diluent to produce concentrations spanning the assay's dynamic range. For the recovery experiment, the samples were spiked with standard solutions of a specific concentration and then serially diluted to 2-fold, 4-fold, 8-fold, and 16-fold dilutions, respectively. The average recovery rate was subsequently calculated.
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse NOV. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse NOV and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse NOV, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Mouse NOV. You can calculate the concentration of NOV in the samples by comparing the OD of the samples to the standard curve.
NOV (CCN3) is a secreted matricellular protein belonging to the conserved CCN family, featuring four typical modular domains: IGFBP, vWF type C, thrombospondin‑1, and a C‑terminal cysteine knot domain. Human CCN3 shares 80% amino acid identity with its mouse homolog, indicating high cross‑species conservation. Widely expressed during development, CCN3 is abundant in muscle, endothelial cells, nervous tissues, adrenal cortex and chondrocytes. Full‑length CCN3 is secreted into the extracellular matrix, binding integrin αvβ3 and α5β1 to mediate endothelial adhesion, chemotaxis and angiogenesis, and suppresses cell proliferation. It modulates hematopoietic, osteogenic and chondrogenic cell self‑renewal and maturation via Notch and BMP signaling, and interacts with multiple partners including Notch1, CCN2, fibulin 1C, Connexin43 and S100A4. Truncated nuclear CCN3 functions as a transcriptional repressor to promote cell proliferation. It also inhibits myogenic gene expression and is closely associated with the progression of Wilm’s tumor and nephroblastoma.
