This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to SEMA4D . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for SEMA4D and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain SEMA4D , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 14 nm. The OD value is proportional to the concentration of SEMA4D . You can calculate the concentration of SEMA4D in the samples by comparing the OD of the samples to the standard curve.
SEMA4D, also known as semaphorin 4D, is a gene located on human chromosome 9q22.2, encoding a molecule with a protein product. This gene belongs to several important protein families, including the CD molecule family, the immunoglobulin-like domain-containing family, and the semaphorins family. Additionally, the SEMA4D gene has an alternative name, CD100, which reflects its crucial role in intercellular communication and immune responses. SEMA4D is a transmembrane protein, consisting of an extracellular region, a transmembrane region, and a cytoplasmic region. Its extracellular region contains a Semaphorin domain, which is used to interact with other proteins and trigger signal transduction. SEMA4D plays a key role in neural development. It is involved in the guidance of nerve axons and the formation of synapses. Through its interaction with receptors (such as Plexin family receptors), SEMA4D can influence the growth and connection of neurons. SEMA4D also has an important role in the immune system. It can regulate the activation and migration of immune cells. The interaction between SEMA4D and its receptors (such as CD72 and PlexinB2) can affect the functions of T cells, B cells, and macrophages. SEMA4D is a multifunctional molecule that plays a role in neural development, the immune system, and diseases. Studying the function and regulatory mechanisms of SEMA4D can help us better understand the regulation of neural development and immune responses, and also contribute to the development of new treatment methods, especially in the field of cancer. The SEMA4D protein belongs to the semaphorins family, and proteins in this family usually play a key role in intercellular communication. Specifically, SEMA4D stands out in the following aspects: Immune response: SEMA4D helps regulate the activity of immune cells, ensuring that the immune system can effectively combat foreign pathogens. Neural development: In the nervous system, SEMA4D participates in guiding the growth and connection of nerve cells, which is crucial for the normal development of the brain. Cancer: Studies have found that SEMA4D is abnormally expressed in some cancers, which may affect tumor growth and spread. Understanding the status of the SEMA4D gene can help us better manage health. For example, if a mutation in the SEMA4D gene is detected, it may indicate potential problems with the individual's immune system function or special needs in neural development. Additionally, for cancer patients, the status of the SEMA4D gene can provide important information about disease progression and treatment response.
