Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse VCAM-1. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse VCAM-1 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse VCAM-1, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 16 nm. The OD value is proportional to the concentration of Mouse VCAM-1. You can calculate the concentration of Mouse VCAM-1 in the samples by comparing the OD of the samples to the standard curve.
Background
Vascular cell adhesion molecule-1 (VCAM-1, CD106) is a type I membrane glycoprotein of the immunoglobulin superfamily. Mouse VCAM-1 has two isoforms: a 110 kDa transmembrane long isoform and a 45 kDa GPI-anchored short isoform, both releasable as soluble VCAM-1. VCAM-1 is induced on vascular endothelium by inflammatory stimuli and constitutively expressed on non-vascular cells including dendritic cells, macrophages, fibroblasts, bone marrow stromal cells, and others. It mediates cell adhesion and signal transduction by binding to ligands such as integrin α4β1, α4β7, αDβ2, and α9β1. VCAM-1 is critical for leukocyte recruitment to inflammation sites, T/B cell development, hematopoietic progenitor cell homing, and may facilitate melanoma metastasis. Soluble VCAM-1 promotes angiogenesis and chemotaxis. It is also associated with pathological conditions like atherosclerosis, rheumatoid arthritis, systemic lupus erythematosus, inflammatory bowel disease, and allograft rejection.
