Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to AChE . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for AChE and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain AChE , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of AChE . You can calculate the concentration of AChE in the samples by comparing the OD of the samples to the standard curve.
Background
Acetylcholinesterase (ACHE) is a member of the alpha/beta hydro lase superfamily. It is a serine hydrolase with a carboxyl esterase activity. Its main function is to terminate transmission at cholinergic synapses by hydrolyzing the neurotransmitter acetylcholine (ACH). Its inhibitors,such as donepezil,rivastigmine,and galantamine,have been approved to treat mild-to-moderate Alzheimer's disease and other dementia. Although ACHE is an essential enzyme primarily expressed in the central and peripheral nervous systems,its expression has been observed in various cell types including fibroblasts,osteoblasts,erythrocytes,vascular endothelial cells,and leukocytes. In addition to the main function,its role includes neurogenesis,neurodegeneration,apoptotic sensitivity,cellular proliferation and differentiation,and a possible role on tumorigenesis.
