Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to PDGF-BB . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for PDGF-BB and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain PDGF-BB , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of PDGF-BB . You can calculate the concentration of PDGF-BB in the samples by comparing the OD of the samples to the standard curve.
Background
PDGF-BB is a mitogen that was initially identified as simian sarcoma viral oncogene homolog. It belongs to the Platelet-Derived Growth Factor (PDGF) family which includes five members: PDGF-AA,PDGF-BB,PDGF-AB,PDGF-CC,and PDGF-DD. PDGF-AA,AB,and BB dimers are processed intracellularly and secreted as active dimers that readily activate PDGF receptors (PDGFRs). PDGF-BB exerts its biological functions through the activation of dimeric receptors that are made up of two structurally similar protein-tyrosine kinase receptor subunits (αα-,αβ-,or ββ-PDGFR). PDGF-BB induces VSMC dedifferentiation by the down regulation of contractile protein markers and the up regulation of MMP-2. In cutaneous remodeling,studies ex vivo revealed that PDGF-BB is a mitogen and chemoattractant for dermal fibroblasts. Abnormalities of PDGFR/PDGF are thought to contribute to a number of human diseases,especially malignancy. Autocrine signaling is a consequence of PDGF-BB overexpression,which is clearly implicated in the pathogenesis of dermatofibrosarcoma protruberans (DFSP).
