Performance
| Typical data |
The following data was generated by the Quality Control Department, under controlled laboratory conditions (ambient temperature: 18-25 °C, relative humidity: 35-75%) using standardized procedures (TMB reaction at 37 °C in the dark for 15 minutes, followed by termination and OD measurement). These values are provided for reference only.
Actual results may vary due to differences in laboratory conditions, operator technique, and equipment. Users are required to generate a standard curve using their own experimental data.

| Precision |
Intra-assay Precision (Within-run Precision): Three samples representing low, mid, and high concentrations of Human ICAM-1 were tested 20 times on a single plate.
Inter-assay Precision (Between-run Precision): Three samples representing low, mid, and high concentrations of Human ICAM-1 were tested on three separate plates, with 20 replicates per plate, to assess variability among assays.

| Recovery |
The recovery of Human ICAM-1 was evaluated by spiking samples at low, mid, and high concentrations across the assay range in various sample matrices.
The assay performance was assessed by comparing the measured concentrations to the expected spiked amounts to determine the percent recovery.
|
|||||||||||||||||||||||||||||||||||||||||||||
| Linearity |
Linearity of the assay was evaluated by spiking samples with high concentrations of Human ICAM-1 and performing serial dilutions using Standard & Sample Diluent to produce concentrations spanning the assay's dynamic range.
The measured values were then compared to the expected concentrations to assess the linearity of response.

| Stability |
Each kit batch is subjected to accelerated stability testing and real-time stability monitoring. Sample performance is evaluated after storage at 37 °C for 10 days to assess the impact of elevated temperature on assay reliability and reagent integrity.

Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human ICAM-1/CD54. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human ICAM-1/CD54 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human ICAM-1/CD54, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human ICAM-1/CD54. You can calculate the concentration of Human ICAM-1/CD54 in the samples by comparing the OD of the samples to the standard curve.
Background
Human intercellular adhesion molecule 1 (ICAM-1), also designated CD54, is a transmembrane glycoprotein of the immunoglobulin superfamily with an apparent molecular mass of approximately 90 kDa. It contains five immunoglobulin-like extracellular domains, a single transmembrane segment, and a short cytoplasmic tail. ICAM-1 is constitutively expressed at low levels on endothelial cells, leukocytes, fibroblasts, and epithelial cells and is strongly upregulated by pro-inflammatory cytokines such as interleukin-1 and tumor necrosis factor alpha.
ICAM-1 binds the leukocyte integrins lymphocyte function-associated antigen 1 (LFA-1) and macrophage-1 antigen (Mac-1), and this interaction is the critical step converting leukocyte rolling into firm adhesion and subsequent transendothelial migration. Through these engagements ICAM-1 governs leukocyte recruitment to inflamed tissue, supports immunological synapse formation and T cell activation, and serves as the principal entry receptor for rhinovirus and as an adhesion receptor for Plasmodium falciparum-infected erythrocytes.
A soluble form generated by proteolytic shedding reflects endothelial activation and systemic inflammation and is associated with cardiovascular disease, autoimmune disease, and tumor progression. This ELISA kit enables quantitative determination of human ICAM-1 in serum, plasma, and cell culture supernatant, supporting research on inflammation, leukocyte trafficking, endothelial biology, and vascular disease.
