Precision

Intra-assay Precision (Within-run Precision): Three samples representing low, mid, and high concentrations of Human IL-6 were tested 20 times on a single plate.

Recovery

The recovery of Human IL-17F was evaluated by spiking samples at low, mid, and high concentrations across the assay range in various sample matrices.

Linearity

Linearity of the assay was evaluated by spiking samples with high concentrations of Human IL-17F and performing serial dilutions using Standard & Sample Diluent to produce concentrations spanning the assay's dynamic range. Test Principle

This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human IL-17F. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human IL-17F and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human IL-17F, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human IL-17F. You can calculate the concentration of Human IL-17F in the samples by comparing the OD of the samples to the standard curve.

Background

L-17F belongs to the IL-17 cytokine family which includes IL-17A, B, C, D, and E (also called IL-25). Similar to IL-17, IL-17F utilizes IL-17RA and IL-17RC as its receptor and employs Act1 and TRAF6 as its signal transducers to induce the expression of pro-inflammatory cytokines and chemokines in many different cell types. IL-17F expression is upregulated in inflammatory bowel disease. A His161 to Arg161 (H161R) substitution in the third exon of the IL17F gene seems to be associated with asthma and chronic obstructive pulmonary disease (COPD) in Japanese subjects. In addition polymorphism of IL-17F seems to be associated to susceptibility to gastric cancer.
