Performance
| Typical data |
The following data was generated by the Quality Control Department, under controlled laboratory conditions (ambient temperature: 18-25 °C, relative humidity: 35-75%) using standardized procedures (TMB reaction at 37 °C in the dark for 15 minutes, followed by termination and OD measurement). These values are provided for reference only.
Actual results may vary due to differences in laboratory conditions, operator technique, and equipment. Users are required to generate a standard curve using their own experimental data.

| Precision |
Intra-assay Precision (Within-run Precision): Three samples representing low, mid, and high concentrations of Human IL-8 were tested 20 times on a single plate.
Inter-assay Precision (Between-run Precision): Three samples representing low, mid, and high concentrations of Human IL-8 were tested on three separate plates, with 20 replicates per plate, to assess variability among assays.

| Recovery |
The recovery of Human IL-8 was evaluated by spiking samples at low, mid, and high concentrations across the assay range in various sample matrices.
The assay performance was assessed by comparing the measured concentrations to the expected spiked amounts to determine the percent recovery.
|
|||||||||||||||||||||||||||||||||||||||||||||
| Linearity |
Linearity of the assay was evaluated by spiking samples with high concentrations of Human IL-8 and performing serial dilutions using Standard & Sample Diluent to produce concentrations spanning the assay's dynamic range.
The measured values were then compared to the expected concentrations to assess the linearity of response.

| Stability |
Each kit batch is subjected to accelerated stability testing and real-time stability monitoring. Sample performance is evaluated after storage at 37 °C for 10 days to assess the impact of elevated temperature on assay reliability and reagent integrity.

Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human IL-8. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Human IL-8 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Human IL-8, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human IL-8. You can calculate the concentration of Human IL-8 in the samples by comparing the OD of the samples to the standard curve.
Background
Human interleukin 8 (IL-8), also termed GCP-1 or NAP-1 and now designated CXCL8, is an 8-9 kDa heparin-binding member of the ELR-positive CXC chemokine subfamily. Monocytes, neutrophils, fibroblasts, keratinocytes, mast cells, smooth muscle cells, dendritic cells, alveolar type II cells and endothelial cells all secrete it upon stimulation by pro-inflammatory cytokines or microbial products, and it is the principal chemoattractant for neutrophils.
IL-8 signals through the G protein-coupled receptors CXCR1 and CXCR2. CXCR2, engaged at low concentrations, mediates chemotaxis and MMP-9 release, whereas CXCR1 at high concentrations triggers the respiratory burst and antimicrobial responses. Serpin A1, also called alpha-1 antitrypsin, blocks CXCR1 by forming a complex with the chemokine, and proteolytic processing provides additional regulation of activity.
Beyond acute inflammation, IL-8 promotes angiogenesis, atherosclerosis and cancer progression by inducing vascular endothelial growth factor, supporting endothelial growth and facilitating tumor invasion and metastasis. As the principal neutrophil chemoattractant it is a key mediator of tissue damage in many inflammatory diseases. This ELISA kit enables quantitative determination of IL-8 in blood, lavage fluid and other specimens, supporting inflammation and cancer research.
