Detection principle

When cells undergo apoptosis, specific DNA endonucleases will be activated, cutting the genomic DNA between the nucleosomes. The DNA of apoptotic cells is cleaved into multimers of 180~200bp fragments, corresponding to the oligonucleosomal size. Therefore, the DNA of apoptotic cells typically migrates as a ladder of 180~200bp on an agarose gel. The exposed 3'-OH of the broken DNA can be catalyzed by Terminal Deoxynucleotidyl Transferase (TdT) with fluorescein labeled dUTP, which can be detected with fluorescence microscope.

Asked: Hi, myself Dr. Shyma K. Latheef, working as Scientist in the Indian Veterinary Research Institute. We have purchased E-CK-A320 One-step TUNEL Apoptosis Kit (Green, FITC) 50 Assays - LOT no. AK13198. Since we have almost 50 test samples to check, before proceeding with our actual samples, it is requested to provide a sample kit of lesser reactions (min. 10 assays) , so that we can standardize the protocol from our end. Expecting a favorable response from your end. Thanks & Regards

Asked: Hi, I am using the one step TUNEL Apoptosis Kit (Green, FITC) for my samples. I want to know about the positive and the negative controls used in this assay kit. Is the POSITIVE CONTROL the one I used in my experiment as treated positive control group? Same is with the NEGATIVE CONTROL is it the one I used in my study as negative control group? I request you to please clarify it as early as possible. Thank you
