This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human IgA. Samples (or Standards) and Horseradish Peroxidase (HRP) linked antibody specific for Human IgA are added to the micro ELISA plate wells. Human IgA in samples (or standards) combines with the coated antibody and HRP linked detection antibody special to Human IgA.Excess conjugate and unbound sample or standard are washed from the plate. The substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450±2 nm. The OD value is proportional to the concentration of Human IgA. The concentration of Human IgA in the samples is then determined by comparing the OD of the samples to the standard curve.
Immunoglobulin A (IgA) is divided into two types: serotype and secretory type.Serotype IgA is a monomer, its content in normal human serum is second only to IgG, accounting for 10% to 20% of the total serum Ig, and its immune effect is relatively weak.Secretory IgA comes in two forms and three forms. It is the main component of the body's mucosal defense system and is widely distributed in milk, saliva, and mucosal secretions of the gastrointestinal tract, respiratory tract, and genitourinary tract.It can inhibit the attachment of microorganisms to the respiratory epithelium, slow down the reproduction of viruses, and has an important immune barrier function. It has antibody activity against certain viruses, bacteria and general antigens, and is the first line of defense to prevent pathogens from invading the body.IgA cannot cross the placenta.There are no IgA antibodies in newborn serum, but secretory IgA can be obtained from breast milk.IgA can appear in the blood of newborns 4 to 6 months after birth, and then gradually increases, reaching a peak in adolescence.
