Test Principle
This ELISA kit uses the Competitive-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with Universal VMA. During the reaction, Universal VMA in the sample or standard competes with a fixed amount of Universal VMA on the solid phase supporter for sites on the Biotinylated Detection Ab specific to Universal VMA. Excess conjugate and unbound sample or standard are washed away, and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns from blue to yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of Universal VMA in tested samples can be calculated by comparing the OD of the samples to the standard curve.
Background
Vanillylmandelic Acid (VMA for short) is a chemical also known as vanillylmandelic acid or 3-methoxy-4-hydroxymandelic acid. It is one of the end products of the metabolism of adrenaline and norepinephrine in the body.
The production process of VMA in vivo mainly involves the action of catechol methyl-converting enzyme and monoamine oxidase. Adrenaline and norepinephrine are catalyzed by these enzymes and are eventually converted into VMA and excreted in the urine.
The detection of VMA has certain application value in medicine. For example, by measuring the amount of VMA excretion in the urine, it can help doctors diagnose certain diseases, such as pheochromocytoma, sympathetic neuroblastoma, etc. In addition, VMA can also serve as a marker of neurotransmitter metabolic disorders, as well as have antioxidant activity, capable of scavenging DPPH free radicals.
