Test Principle
This ELISA kit uses the Competitive-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with Universal 5-HIAA. During the reaction, Universal 5-HIAA in the sample or standard competes with a fixed amount of Universal 5-HIAA on the solid phase supporter for sites on the Biotinylated Detection Ab specific to Universal 5-HIAA. Excess conjugate and unbound sample or standard are washed away, and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns from blue to yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of Universal 5-HIAA in tested samples can be calculated by comparing the OD of the samples to the standard curve.
Background
5-hydroxyindoleacid (5-Hydroxyindoleacetic Acid, 5-HIAA for short) is a kind of important bioactive substances, which can be found widely in organisms and play an important role in the nervous system and endocrine system.
5-HIAA is an important intermediate product in tryptophan metabolism pathway, which is produced by tryptophan through a series of enzymatic reactions. It is one of the main metabolites of serotonin (5-hydroxytryptamine), an important neurotransmitter involved in regulating mood, sleep, appetite and many other physiological functions.
In the medical field, 5-HIAA detection is widely used in the diagnosis and treatment of a variety of diseases. For example, by detecting 5-HIAA levels in urine or blood, it can assist in the diagnosis of tumor diseases such as carcinoid syndrome and neuroblastoma. In addition, 5-HIAA can also be used to evaluate the treatment effect of depression, anxiety and other mental disorders.
