This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to TGF-β1. Samples (or Standards &Quality Control Material) are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for TGF-β1 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain TGF-β1, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450±2 nm. The OD value is proportional to the concentration of TGF-β1. You can calculate the concentration of TGF-β1 in the samples by comparing the OD of the samples to the standard curve.
TGFB1 (TGF-β1) is a 44 kDa multifunctional cytokine regulating cell growth, differentiation, migration, immunity, and development. It exists as a latent proprotein, where the Latency-Associated Peptide (LAP) non-covalently binds TGF-β1, maintaining inactivity and storing it in the extracellular matrix. Activation is tightly controlled via interactions of LAP with milieu proteins (LTBP1, GARP, NRROS) and specific integrins (αvβ6/αvβ8), which induce conformational release of active TGF-β1.Active TGF-β1 signals through TGFBR1/TGFBR2 receptors, inducing SMAD2/3 phosphorylation and regulating gene expression. It modulates immune responses, directing T-cell differentiation toward Treg or Th17 lineages in a concentration-dependent manner. It is crucial for wound healing, bone remodeling, and organogenesis, but its dysregulation is linked to fibrosis, cancer progression, metastasis, atherosclerosis, and autoimmune diseases.
