Test Principle
This ELISA kit uses the Competitive-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with Universal LXA4. During the reaction, Universal LXA4 in the sample or standard competes with a fixed amount of Universal LXA4 on the solid phase supporter for sites on the Biotinylated Detection Ab specific to Universal LXA4. Excess conjugate and unbound sample or standard are washed away, and Avidin-Horseradish Peroxidase (HRP) conjugate are added to each micro plate well and incubated. Then a TMB substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns from blue to yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of Universal LXA4 in tested samples can be calculated by comparing the OD of the samples to the standard curve.
Background
Lipoxin A4 (LXA4) is a trihydroxy fatty acid containing a conjugated tetraene, produced by the metabolism of 15-HETE or 15-HpETE with human leukocytes. LXA4 is equipotent to leukotriene B4 (LTB4; Item Nos. 10007240 | 20110) in inducing superoxide generation in human neutrophils at 0.1 μM. LXA4 is associated with several other biological functions including leukocyte activation, chemotaxis effects, natural killer cell inhibition, and monocyte migration and adhesion.
