Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to CXCL1 . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for CXCL1 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain CXCL1 , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 37 nm. The OD value is proportional to the concentration of CXCL1 . You can calculate the concentration of CXCL1 in the samples by comparing the OD of the samples to the standard curve.
Background
CXCL1/ GROα, also known as KC in mice and cinc1 in rats, is a member of the CXC chemokine family. Human CXCL1/ GROα has a total length of 107 amino acids (aa) with an estimated molecular weight of 11 kDa. The sequence homology of mouse and rat homologues to human protein was 57% and 59%, respectively. Genome-wide analysis showed that the protein encoded by the mouse Cxcl3 gene was homologous to the human CXCL1/ GROα protein. Human CXCL1/ GROα can be cleaved into three isoforms: CXCL1/ GROα (aa 4-73), CXCL1/ GROα (aa 5-73) and CXCL1/ GROα (aa 6-73), all of which are more active than full-length proteins. CXCL1/ GROα is secreted by mast cells and macrophages, so it acts as a chemical attractant of neutrophils; GRO-α plays an important role in inflammation and immune response, enhances immune defense by attracting and activating immune cells, is up-regulated in the tumor microenvironment, and is closely related to tumor growth, angiogenesis and metastasis. It can promote tumor progression by activating tumor-associated fibroblasts and endothelial cells.
