Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to ESM1 . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for ESM1 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain ESM1 , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 31 nm. The OD value is proportional to the concentration of ESM1 . You can calculate the concentration of ESM1 in the samples by comparing the OD of the samples to the standard curve.
Background
Endothelial cells are multifunctional cells that cover the inner surface of blood vessels and regulate a variety of important physiological and pathological reactions. Endothelial cells are dynamic participants in cell and organ function, rather than a passive barrier. Through the expression of surface proteins and the secretion of soluble mediators, endothelial cells control vascular tone and permeability, regulate coagulation and thrombosis, and direct the recruitment and homing of white blood cells to specific areas of inflammation. This endothelial cell dependent control of white blood cell roll, adhesion, and transendothelial migration involves several known endothelial cell surface molecules, including E- and P-selectin, ICAMs, and VCAM-1, which are differentially regulated by pro-inflammatory cytokines such as TNF· and IFNA.
