Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to MMP-9 . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for MMP-9 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain MMP-9 , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 26 nm. The OD value is proportional to the concentration of MMP-9 . You can calculate the concentration of MMP-9 in the samples by comparing the OD of the samples to the standard curve.
Background
The full name of MMP-9 in Chinese is matrix metalloproteinase 9. The MMP-9 gene is located on chromosome 20q11.1-13.1, 26-27kpp, with 13 exons and 9 introns. It belongs to the matrix metalloproteinase (MMP) family. The main function of MMP-9 is to degrade and reshape the dynamic balance of extracellular matrix. The homology of MMP-9 mRNA in mammals is about 80%. MMP-9 can decompose structural complexes such as ECM and basement membrane in the respiratory tract and lungs, thus participating in the reconstruction of the respiratory tract and lungs. It can also regulate the activity of other proteases and cytokines. It can degrade alpha 1 antitrypsin, protect neutrophil elastase activity, enhance collagen cell and MMP-13 sol-gel activity in collagen colloids. MMP-9 can also break down a 62 amino acid peptide from interleukin-8 (CXCL8/CL8), increasing its chemotactic activity towards neutrophils by 10 times, but it also inhibits other neutrophil chemotactic factors. In addition, MMP-9 binds to CD44 to release stored TGF - β 1, and MMP-9 can participate in angiogenesis by releasing vascular endothelial growth factor (VEGF).
