Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to VEGF165 . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for VEGF165 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain VEGF165 , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 27 nm. The OD value is proportional to the concentration of VEGF165 . You can calculate the concentration of VEGF165 in the samples by comparing the OD of the samples to the standard curve.
Background
Human VEGF165 (Vascular endothelial growth factor 165) is one of the most important splicing variants in the VEGF-A family, with high biological activity and a wide range of physiological functions. Composed of 165 amino acids, VEGF165 is a dimeric glycoprotein that plays its role in promoting angiogenesis by binding to VEGFR-1 and VEGFR-2 receptors. VEGF165 is an important driver of tumor angiogenesis and is therefore a target for anti-tumor therapy. For example, Bevacizumab is a monoclonal antibody against VEGF-A that inhibits VEGf165-mediated angiogenesis
