Recovery

Linearity
Linearity of the assay was evaluated by spiking samples with high concentrations of Mouse DKK3 and performing serial dilutions using Standard & Sample Diluent to produce concentrations spanning the assay's dynamic range. For the recovery experiment, the samples were spiked with standard solutions of a specific concentration and then serially diluted to 2-fold, 4-fold, 8-fold, and 16-fold dilutions, respectively. The average recovery rate was subsequently calculated.
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse DKK3. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse DKK3 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse DKK3, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 12 nm. The OD value is proportional to the concentration of Mouse DKK3. You can calculate the concentration of Mouse DKK3 in the samples by comparing the OD of the samples to the standard curve.
Dkk3 protein antagonizes canonical Wnt signaling by inhibiting LRP5/6 interaction with Wnt and by forming a ternary complex with the transmembrane protein KREMEN that promotes internalization of LRP5/6. DKKs play an important role in vertebrate development, where they locally inhibit Wnt regulated processes such as antero-posterior axial patterning, limb development, somitogenesis and eye formation. In the adult, Dkks are implicated in bone formation and bone disease, cancer and Alzheimer disease.
