Recovery
The recovery of Mouse TFPI was evaluated by spiking samples at low, mid, and high concentrations across the assay range in various sample matrices.
The assay performance was assessed by comparing the measured concentrations to the expected spiked amounts to determine the percent recovery.

Linearity
Linearity of the assay was evaluated by spiking samples with high concentrations of Mouse TFPI and performing serial dilutions using Standard & Sample Diluent to produce concentrations spanning the assay's dynamic range. The measured values were then compared to the expected concentrations to assess the linearity of response.

For the recovery experiment, the samples were spiked with standard solutions of a specific concentration and then serially diluted to 2-fold, 4-fold, 8-fold, and 16-fold dilutions, respectively. The average recovery rate was subsequently calculated.
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse TFPI. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse TFPI and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse TFPI, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 13 nm. The OD value is proportional to the concentration of Mouse TFPI. You can calculate the concentration of Mouse TFPI in the samples by comparing the OD of the samples to the standard curve.
Tissue Factor Pathway Inhibitor (TFPI), also called extrinsic pathway inhibitor (EPI) and lipoprotein-associated coagulation inhibitor (LACI), is a key Kunitz-type serine protease inhibitor regulating the extrinsic blood coagulation pathway. It directly inhibits factor X (Xa) and, in an Xa-dependent way, inhibits VIIa/tissue factor (TF) activity by forming a quaternary Xa/LACI/VIIa/TF complex, playing a vital antithrombotic role.TFPI is mainly produced by vascular endothelial cells; it is also expressed by vascular smooth muscle cells, megakaryocytes/platelets, monocytes and other cells, and detected in macrophages and T cells in atherosclerotic lesions. Its structure has three tandem Kunitz domains (K1, K2, K3) and a basic C-terminal region: K2 inhibits FXa’s active site, while K1 inhibits FVIIa’s to block the TF–FVIIa complex’s procoagulant activity.
