Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to TRACP-5b . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for TRACP-5b and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain TRACP-5b , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 42 nm. The OD value is proportional to the concentration of TRACP-5b . You can calculate the concentration of TRACP-5b in the samples by comparing the OD of the samples to the standard curve.

Tartrate-resistant acid phosphatases (TRAPIACPS) are metallic enzymes belonging to the acid phosphatase group that catalyze the conversion of orthophosphate monomers into alcohols and orthophosphates.It is a ferriglycoprotein synthesized with a 35kd monomeric proenzyme.ACP5 is involved in bone resorption and is secreted by active osteoclasts.1 Dephosphorylation of osteopontin, a bone matrix protein, subsequently promotes osteoclast detachment and migration.In colorectal cancer, ovarian cancer, breast cancer, melanoma and other cancers,ACP5 expression is associated with tumor progression.In vitro experiments have shown that down-regulated expression of ACP5 can promote the proliferation and invasion of tumor cells in colorectal cancer cell lines.In senum, ACPS has been used as a marker for thyroid metastasis, and in addition, Acps removes the lysosomal protein as a recognition marker for mannose 6-phosphate.Tartrate-resistant acid phosphatases (TRAPIACPS) are metallic enzymes belonging to the acid phosphatase group that catalyze the conversion of orthophosphate monomers into alcohols and orthophosphates.
