Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to CXCL2 . Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for CXCL2 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain CXCL2 , biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 52 nm. The OD value is proportional to the concentration of CXCL2 . You can calculate the concentration of CXCL2 in the samples by comparing the OD of the samples to the standard curve.
Background
CXCL2/GRO beta, also called MIP-2 in mouse and CINC-3 in rat, is a member of the CXC chemokine family. Human CXCL2/GRO beta is 107 amino acids (aa) in length with a predicted molecular weight of 11 kDa. The mouse and rat orthologs share 70% and 71% aa sequence identity with the human protein, respectively. N-terminal aa 1-4 of CXCL2/GRO beta can be post-translationally cleaved which confers enhanced hematopoietic bioactivity. CXCL2/GRO beta is produced by a variety of cell types including monocytes and macrophages at sites of inflammation and is chemotactic for granulocytes, including neutrophils.
