Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Mouse FcεRⅡ/CD23. Standards or samples are added to the micro ELISA plate wells and combined with the specific antibody. Then a biotinylated detection antibody specific for Mouse FcεRⅡ/CD23 and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Mouse FcεRⅡ/CD23, biotinylated detection antibody and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Mouse FcεRⅡ/CD23. You can calculate the concentration of Mouse FcεRⅡ/CD23 in the samples by comparing the OD of the samples to the standard curve.
Background
CD23 is a 45 kD protein, also known as Leu-20, FcεRII, IgE Fc receptor, BLAST-2, B6, and low affinity IgE receptor. CD23 responds to high levels of IgE by downregulating IgE secretion. In human monocytes, CD23 triggering results in release of pro-inflammatory cytokines including TNF-α, IL-1, IL-6, and GM-CSF. CD23 can be proteolytically cleaved to generate soluble CD23 fragments of various molecular weights. In chronic lymphocytic leukemia, levels of soluble CD23 in the serum can be used as a prognostic marker to identify patients at high risk for disease progression.
