Test Principle
This ELISA kit uses the Competitive-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with Canine Cortisol. Samples (or Standards) and Horseradish Peroxidase (HRP) linked antibody specific for Canine Cortisol are added to the micro ELISA plate wells. Canine Cortisol in samples (or standards) competes with a fixed amount of Cortisol on the solid phase supporter for sites on the HRP linked detection antibody specific to Cortisol. Excess conjugate and unbound sample or standard are washed from the plate. The substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450±2 nm. The concentration of Canine Cortisol in the samples is then determined by comparing the OD of the samples to the standard curve.

Cortisol is a steroid hormone released from the adrenal cortex inresponse to the hormone ACTH (produced by the pituitary gland)Cortisol is involved in the response to stress; it increases blood pressure,blood sugar levels, suppresses the immune system and may causeinfertility in women. Cortisol acts through specific intracellular receptorsand has effects in numerous physiologic systems, including immunefunction, glucose-counter regulation, vascular tone, substrate utilizationand bone metabolism. Cortisol is excreted primarily in urine in anunbound (free) form. Cortisol is bound with high affinity in plasma tocorticosteroid-binding globulin (CBG, transcotin) and to albumin. Onlyfree Cortisol is available to most receptors.
