Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human CA19-9. Samples (or Standards) and Horseradish Peroxidase (HRP) linked antibody specific for Human CA19-9 are added to the micro ELISA plate wells. Human CA19-9 in samples (or standards) combines with the coated antibody and HRP linked detection antibody special to Human CA19-9.Excess conjugate and unbound sample or standard are washed from the plate. The substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human CA19-9. The concentration of Human CA19-9 in the samples is then determined by comparing the OD of the samples to the standard curve.
Background
Mucin glycoprotein is a Sialyl Lewis A structure which is synthesized from type 1 blood group precursor chains and is present in individuals expressing the Lewis A and/or Lewis B blood group antigens. In normal tissues, sialyl Lewis A antigen is present in ductal epithelium of the breast, kidney, salivary gland, and sweat glands. Its expression is greatly enhanced in serum as well as in the majority of tumor cells in gastrointestinal (GI) carcinomas, including adenocarcinomas of the stomach, intestine, and pancreas. Preoperative elevated CA19-9 levels in patients with stage I pancreatic carcinoma decrease to normal values following surgery. When used serially, CA19-9 can predict recurrence of disease prior to radiographic or clinical findings.
