This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human IgG. Samples (or Standards) and Horseradish Peroxidase (HRP) linked antibody specific for Human IgG are added to the micro ELISA plate wells. Human IgG in samples (or standards) combines with the coated antibody and HRP linked detection antibody special to Human IgG.Excess conjugate and unbound sample or standard are washed from the plate. The substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human IgG. The concentration of Human IgG in the samples is then determined by comparing the OD of the samples to the standard curve.
IgG is a monomeric immunoglobulin composed of two identical heavy chains and two identical light chains linked together by inter-chain disulfide bonds. There are four IgG subclasses (IgG1, IgG2, IgG3, and IgG4) in humans named in order of decreasing abundance. They differ in the size of the hinge region and the number and arrangements of the interchain disulfide bonds linking their heavy chains. Of the 5 immunoglobulin isotypes, IgG is most abundant in human serum. IgG is synthesized and secreted by plasma B cells and is involved in the secondary immune response. The measurement of IgG can be a diagnostic tool for certain conditions such as autoimmune diseases, gastrointestinal conditions or immunoglobulin-deficiency disorders.
