Test Principle
This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human LDLR. Samples (or Standards) and Horseradish Peroxidase (HRP) linked antibody specific for Human LDLR are added to the micro ELISA plate wells. Human LDLR in samples (or standards) combines with the coated antibody and HRP linked detection antibody special to Human LDLR.Excess conjugate and unbound sample or standard are washed from the plate. The substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human LDLR. The concentration of Human LDLR in the samples is then determined by comparing the OD of the samples to the standard curve.
Background
LDLR (low density lipoprotein receptor) is a member of the LDL receptor gene family and is involved in receptor-mediated endocytosis of specific ligands. The LDLR is a cell surface glycoprotein that scavenges LDL from the blood and regulates plasma LDL cholesterol. The cytoplasmic domain of the LDL receptor is necessary for the receptor to cluster in coated pits,which promotes the rapid endocytosis of bound LDL. The protein is highly glycosylated through N- and O-linkages and thus migrates at 100 to 160 kDa bands on SDS-PAGE.
