This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antibody specific to Human OC. Samples (or Standards) and Horseradish Peroxidase (HRP) linked antibody specific for Human OC are added to the micro ELISA plate wells. Human OC in samples (or standards) combines with the coated antibody and HRP linked detection antibody special to Human OC.Excess conjugate and unbound sample or standard are washed from the plate. The substrate solution is added to each well. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Human OC. The concentration of Human OC in the samples is then determined by comparing the OD of the samples to the standard curve.
Bone gamma-carboxyglutamic acid (Gla) protein, known as BGLAP, BGP or osteocalcin, is an abundant, non-collagenous protein component of bone that is produced by osteoblasts. In mice, osteocalcin is composed of a cluster of 3 genes known as OG1, OG2 and ORG, all of which can be found within a 23Kb span of genomic DNA. Human osteocalcin is a highly conserved, 46-50 amino acid, single chain protein that contains three vitamin K-dependent g-carboxyglutamic acid residues. Osteocalcin appears transiently in embryonic bone at the time of mineral deposition, where it binds to hydroxyapatite in a calcium-dependent manner.
